PCR Amplicon Sequencing

Sequence PCR-amplified DNA using our PCR Amplicon Sequencing service.

This service is recommended for researchers who:

  • Wish to multiplex up to 96 samples to reduce price per sample
  • Want to achieve modal raw read accuracy of Q20+ (99%) and above
  • Want to obtain duplex data
  • Want to optimise their sequencing experiment for accuracy and output
  • Need a PCR-free method of multiplexing to preserve additional information such as base modifications
  • Require control over read length
  • Would like to utilise upstream processes such as size selection or whole genome amplification

Based on the Native Barcoding Kit 96 V14 (Oxford Nanopore Technologies)

This service yields:

  • PCR amplicon sequence

Price is determined based on number of samples provided at a time, referred to as batch size.

FAQ

How much DNA should I send?

Please send 500 fmol of amplicon DNA in 100 µl of water. The necessary weight can be calculated using the equation: amount to send (ng) = 0.308 * amplicon length (bp)

What QC metrics do my samples need to meet?

We run key QC analyses on all received samples. However, to minimize lost time and unnecessary shipping and quality control charges, we recommend completing the following before sending your sample:

  • Perform PCR clean up
  • Ensure sample concentration of 5 fmol/uL by Qubit or similar analysis
  • Confirm 260/280 ratio of 1.8 - 2.0 and 260/230 ratio of 1.9 - 2.5
  • Verify sample has a clean band via gel electrophoresis

How do I ship my samples?

Detailed shipping instructions will be provided via email once your order has been placed:

  1. Samples will need to be loaded in a 96-well plate in the plate position indicated during the ordering process.
  2. Your plate(s) will need to be labeled with your Order ID number.
  3. Prepaid shipping label(s) will be provided to you to print.
  4. Samples will need to be shipped with 10 lbs of dry ice.

How do I track my samples?

The status of your samples can be found within your Wasatch BioLabs portal, where you will be able to view both individual and/or batch sample status. You can also opt-in to email notifications in the app to receive updates in real-time.

How long will it take to get my data after mailing my samples?

Your samples will arrive at the lab within 2 days of you sending them via USPS Priority Express Shipping. Once received, you will have access to the sample data within 5-7 days via your portal.

How will my data be delivered?

Sample data can be directly downloaded from your WBL portal in .fastq format. You can also set up auto-delivery to receive your data via email once it is ready.

Do I need to remove primers from my sample?

Yes. Please perform a PCR cleanup to remove primers, buffers, spare nucleotides, and proteins following the PCR.

What data will I receive?

Results will be returned in .fastq format—the industry standard for sequencing data. To support download performance, the data is split across multiple files and gzipped. For more information on .fastq format and how to combine files, email us at support@wasatchbiolabs.com.
