Targeted DNA Methylation Sequencing

Sequence thousands of genomic DNA (gDNA) regions specific to your research needs with our Targeted DNA Methylation Sequencing service. Combining our proprietary targeted sequencing technology with Oxford Nanopore sequencing, our service yields native DNA sequence as well as methylation and hydroxymethylation data.

This service is recommended for researchers who:

  • Want to sequence and determine methylation status at thousands of regions
  • Want to achieve modal raw read accuracy of Q20+ (99%) and above
  • Want to obtain duplex data
  • Want to optimize their sequencing experiment for accuracy and output
  • Require control over read length
  • Would like to utilize upstream processes such as size selection or whole genome amplification

Based on the Ligation Sequencing Kit V14 (Oxford Nanopore Technologies)

This service yields:

  • Targeted, native DNA sequence and methylation and hydroxymethylation status for thousands of regions
  • Average target enrichment between 400-2,300x
  • Scalable coverage
  • Copy number variation

Price is determined based on number of samples provided at a time, referred to as batch size.

FAQ

How much DNA should I send?

Please send 1 µg (1000 ng) of DNA.

What kind of methylation can this assay detect?

Currently, we can detect 5-methyl and 5-hydroxymethyl cytosines within CpG contexts and are planning on adding additional modifications. Please email us at support@wasatchbiolabs.com if you would like to see other specific epigenetic marks added!

How do I set up a new assay?

We work with you to design a new assay.

  1. You send us a BED format file with the regions you want to target.
  2. Our team of bioinformaticians designs optimized probes to the target regions and sends you a report for final approval.
  3. We handle ordering the probe library and storing it for future runs.

How do I ship my samples?

Detailed shipping instructions will be provided via email once your order has been placed:

  1. Samples will need to be loaded in a 96-well plate in the plate position indicated during the ordering process.
  2. Your plate(s) will need to be labeled with your Order ID number.
  3. Prepaid shipping label(s) will be provided to you to print.
  4. Samples will need to be shipped with 10 lbs of dry ice.

How do I track my samples?

The status of your samples can be found within your Wasatch BioLabs portal, where you will be able to view both individual and/or batch sample status. You can also opt-in to email notifications in the app to receive updates in real-time.

How long will it take to get my data after mailing my samples?

Your samples will arrive at the lab within 2 days of you sending them via USPS Priority Express Shipping. Once received, you will have access to the sample data within 5-7 days via your portal.

How will my data be delivered?

Sample data can be directly downloaded from your WBL portal in .fastq format. You can also set up auto-delivery to receive your data via email once it is ready.

What coverage can I expect?

While coverage will depend on several factors—the quality and quantity of the sample provided, the number of probes in the library, and the efficiency of each probe—we regularly achieve >30x coverage for genomic DNA samples. Because of its low concentration, yields on cfDNA samples will be lower.

How many loci can I target?

We have successfully enriched for 1-2000 loci.
